Aquaculture en système clos : estimation des coûts de production pour l'élevage du bar et du turbot ArchiMer
Lavenant, M; Paquotte, Philippe.
Two major constraints to aquaculture have raised in most countries : the little availability of convenient sites because of the competition for the use of the coastline and the progressive implementation of regulations to reduce the impact of the farms on the environment. The technique of recirculation, widely used for hatcheries, has been developed to answer these constraints and to get free of the natural variations in order to regulate the résults. The ongrowing experimental facilities have proved good enough results to make it possible to conceive industrial projects for sea-bass and turbot. In first approach, the production costs of these systems are similar to traditional techniques but expected productivity gains due to an increase of zootechnical...

Can turbot, Psetta maxima, be fed with self-feeders? ArchiMer
Burel, Christine; Robin, Jean; Boujard, Thierry.
The capability of turbots to use self-feeders was studied using 3 groups of 100 juveniles. Feeding activity reached a plateau after 20 days. Almost all feeding activity was observed during the photophase, and the acrophase was associated with dawn. The proportion of nocturnal demands was the highest in the replicate with the lowest feed efficiency, suggesting that feed waste was mainly occurring during nocturnal feeding activity. During the last 30 days of the experiment, demands were only rewarded during two periods of 2.5 h.day(-1). Feed efficiency was improved and was homogeneous between the three tanks, but the voluntary feed intake was reduced. It is concluded that turbots are able to use self-feeders but they do not adapt quickly, and they are not...

Characterization and variations of organic parameters in teleost fish endolymph during day-night cycle, starvation and stress conditions ArchiMer
Guibbolini, M; Borelli, G; Mayer Gostan, N; Priouzeau, F; De Pontual, Helene; Allemand, D; Payan, P.
The aim of the present work was to examine the modifications of the organic composition of fish endolymph under environmental conditions (day-night cycle, starvation and Cl-2-Stress) known to modify otolith growth. Endolymph electrophoretic patterns were compared. An antibody raised against the trout otolith organic matrix allowed examining the variations of organic matrix precursors in the endolymph under the above conditions. Western blot analysis showed bands around 60-80 kDa. A 50% decrease of immunolabelling was observed during the night whereas increases were seen after starvation (factor 3) or stress (factor 2) suggesting that these variations could be related to the organic matrix deposit. A factor retarding in vitro CaCO3 crystallization (FRC) was...

Comparative effects of long-term hypoxia on growth, feeding and oxygen consumption in juvenile turbot and European sea bass ArchiMer
Pichavant, Karine; Person, Jeannine; Le Bayon, Nicolas; Severe, Armelle; Le Roux, Annick; Boeuf, Gilles.
When juvenile turbot Scophthulmus maximus and sea bass Dicentrarchus labrax were fed to satiation, growth and food intake were depressed under hypoxia (3.2 +/- 0.3 and 4.5 +/- 0.2 mg O-2 l(-1)). However, no significant difference in growth was observed between fishes maintained in hypoxia and fed to satiation and fishes reared in normoxia (7.4 +/- 0.3 mg O-2 l(-1)) and fed restricted rations (same food intake of fishes at 3.2 mg O-2 l(-1)). Routine oxygen consumption of fishes fed to satiation was higher in normoxia than in hypoxia due to the decrease in food intake in the latter. Of the physiological parameters measured, no significant changes were observed in the two species maintained in hypoxia. This study confirms the significant interaction between...

Effect of dietary inulin and oligosaccharides as prebiotics for weaning turbot, Psetta maxima (Linnaeus, C. 1758) ArchiMer
Mahious, A.s.; Gatesoupe, Joel; Hervy, Magali; Metailler, Robert; Ollevier, F.
Preliminary experiments were undertaken to investigate the effect of dietary inulin (Raftiline ST), oligofructose (Raftilose P95) and lactosucrose on the growth and intestinal bacteria of the marine carnivorous turbot, Psetta maxima. Turbot larvae were weaned on compound diets containing 2% Rafiline ST, 2% Raftilose P95 or 2% lactosucrose; 2% cellulose was the carbon source in the control group. The final mean weight of the group weaned with Raftilose P95 was significantly higher than those observed with the other diets. The bacterial load was highly variable in weaning turbot, especially with respect to the putative Vibrio spp. growing on TCBS agar which, in general, seemed to be dominant. Of the total load of bacterial isolates from turbot weaned on...

Effect of dietary lipid sources on odour-active compounds in muscle of turbot (Psetta maxima) ArchiMer
Serot, Thierry; Regost, Christelle; Prost, Carole; Robin, Jean; Arzel, Jacqueline.
Odour-active compounds in muscle of turbot (Psetta maxima) fed experimental diets containing fish oil (FO), soybean oil (SO) or linseed oil (LO) were investigated by a gas chromatography/olfactometry technique. Thirty-one areas associated with odours were detected in muscle extracts. Among the compounds responsible for these odours, 23 were formed by oxidation of unsaturated fatty acids. Independently of diet, (E)-2-penten-1-ol and (E)-3-hexen-1-ol contribute strongly to the odour of turbot. (E,Z)-2,6-Nonadienal, (E)-2-pentenal and (E,E)-1,3-(Z)-5-octatriene seem to contribute strongly to the odour of turbot fed diets containing high levels of n-3 PUFA (FO and LO groups). Hexanal and decanal show a high detection frequency in turbot fed diets containing...

Effect of temperature, volume of ova batches, and addition of a diluent, an antibiotic, oxygen and a protein inhibitor on short-term storage capacities of turbot, Psetta maxima, ova ArchiMer
Suquet, Marc; Chereguini, Olvido; Omnes, Marie-helene; Rasines, Inmaculada; Normant, Yvon; Souto, Isabel; Quemener, Loic.
The effect of different parameters on shea-term storage capacity of turbot ova was assessed over a 45-h period after ova collection for fertilization rates and over a 9-h period after ova collection for hatching rates. Increasing the volume of ova sampling from 0.5 to 2.5 mt, as well as adding an antibiotic-antimicotic solution or oxygen did not significantly change the storage capacity of ova. Regarding the hatching rates, a higher storage ability was recorded at 8 and 13 degrees C, compared to 3 degrees C. The mean composition of the ovarian fluid was determined (n = 57 spawns). Use of a diluent mimicking the ovarian fluid significantly decreased the storage ability as assessed by the fertilization rates but did not modify the hatching rates. Diluting...

Effects of hypoxia on growth and metabolism of juvenile turbot ArchiMer
Pichavant, Karine; Person, Jeannine.
The effects of hypoxia on growth, feed efficiency, nitrogen excretion, oxygen consumption and metabolism of juvenile turbot (120 g) were studied in a 45-day experiment carried out in sea water at 17.0 +/- 0.5 degrees C and 34.5 ppt salinity. Fish were fed to satiation at O-2-concentrations of 3.5 +/- 0.3, 5.0 +/- 0.3 mg l(-1) (hypoxia) and 7.2 +/- 0.3 mg l(-1) (normoxia). Both feed intake (FI) and growth were significantly lower under hypoxia than under normoxia, with no significant differences being observed between 3.5 and 5.0 mg O-2 l(-1). During the first 2 weeks of the experiment, FI was halved under hypoxic conditions, and there were large differences among treatments in feed conversion ratio (FCR), i.e., it was 3.2, 1.5, and 0.9 in turbot exposed to...

Effects of O-2 supersaturation on metabolism and growth in juvenile turbot (Scophthalmus maximus L.) ArchiMer
Person, Jeannine; Pichavant, Karine; Vacher, Coralie; Le Bayon, Nicolas; Severe, Armelle; Boeuf, Gilles.
Effects of O-2 supersaturation on metabolism and growth were studied in juvenile turbot (Scophthalmus maximus L.). When fish were reared for 30 days in water containing O-2 at 147% or 223% air saturation, there were no significant differences in food intake, growth, food conversion or protein utilization compared to fish exposed to normoxia (100% air saturation in water outlet). Exposure to hyperoxia resulted in increased body fat deposition. Daily rates of O-2 consumption of resting fish were not affected by O-2-concentrations, and there were no significant differences in rates of nitrogenous excretion among fish exposed to the different O-2-concentrations. Turbot tolerated severe hyperoxia, 350% air saturation, for 10 days. There were changes in acid-base...

Effects of repeated hypoxic shocks on growth and metabolism of turbot juveniles ArchiMer
Person, Jeannine; Lacut, Anne-sophie; Le Bayon, Nicolas; Le Roux, Annick; Pichavant, Karine; Quemener, Loic.
Turbot juveniles (45 g) were exposed for 41 d (17 C, 34parts per thousand salinity) to constant normoxic (100-100% air saturation, 100-100) or moderate hypoxic (75-75% air saturation, 75-75) conditions and to repeated hypoxic shocks (20% saturation for 1 h, 5 d per week) from normoxic (100-20% air saturation. 100-20) or moderate hypoxic (75-20% air saturation, 75-20) conditions. A normoxic group was feed restricted (100-FR). Mass increase of 100-100 and 75-75 groups fed to satiation was not significantly different. In comparison, it was significantly lower in the 100-20 and 75-20 groups (NS between the two hypoxic shocks groups). Intermediate results were obtained in the 100-100-FR group. The lowest mass increase under hypoxic shocks was explained by a...

Influence de l'hydrodynamique des bassins sur la survie et la croissance des larves de turbot (Psetta maxima) ArchiMer
Gaignon, Jean-louis; Petton, Bruno; Quemener, Loic.
During larval rearing phase of turbot, a substantial variability in survival and growth is frequently observed. In this paper, we studied the effects of hydrodynamics in larvae rearing tanks on the survival and the growth of fed and starved larvae, using the following conditions: water flows of 600 and 1200 ml.mn(-1), constant and progressive ones, air flows of 10 and 30 ml.mn(-1), downward and upward water circulation, with direction change during the rearing phase. We also studied the interactions between water circulation direction and feeding conditions by (1) shifting the first feeding day (4 occasions, at 8 hours intervals, from 52 to 76 hours after 95% of hatching) and (2) using 2 amounts of preys (which differ by a factor 3). The results show that...

Ionic composition of endolymph in teleosts: Origin and importance of endolymph alkalinity ArchiMer
Payan, P; Kossmann, Hans; Watrin, A; Mayer-gostan, N; Boeuf, Gilles.
Ionic (Na+, K+, Cl-, PO43-, pH), total CO2, total calcium and protein concentrations in the plasma and endolymph of the inner ear were compared in trout Oncorhynchus mykiss and turbot Scophthalmus maximus. In both species, saccular endolymph was characterized by high levels of K+ and total CO2 and in trout by an alkaline pH, The kinetic characteristics of proton secretion across the saccular epithelium of trout were investigated using a titration technique in which isolated saccules were mounted as closed sacs. The rate of proton secretion depends strongly on the pH of the Ringer's solution and secretion stops at a pH below 7.2, Proton secretion is driven by an energy-dependent mechanism involving basolateral ouabain-sensitive Na+/K+ exchangers. Proton...

Long-term effects of the cryopreservation of turbot (Psetta maxima) spermatozoa ArchiMer
Suquet, Marc; Dreanno, Catherine; Petton, Bruno; Normant, Yvon; Omnes, Marie-helene; Billard, Roland.
The survival of turbot eggs and the rearing capacities of larvae stemmed from artificial fertilization practices using frozen-thawed spermatozoa were evaluated. Furthermore, the viability of sperm samples stored during a 9 month period in liquid nitrogen was assessed. No significant difference in the fertilization rate, hatching rate, survival and wet weight of 10-day old larvae were observed using fresh or frozen-thawed spermatozoa. The motility recorded at 10 s and 60 s post-activation and the fertilization capacity of frozen-thawed spermatozoa were not significantly decreased during a 9 month storage period in liquid nitrogen. These results confirm the high quality of the turbot spermatozoa stemmed from the cryopreservation process, allowing their use...

Ressources halieutiques hors quotas du Nord Est Atlantique : bilan des connaissances et analyse de scénarios d'évolution de la gestion ArchiMer
Forest, Andre.
Part of the resources caught in the North Sea, Channel or Atlantic Sea do not fall under the « TAC and Quotas » strand of the Common Fisheries Policy, but the European Union may propose that such measures may eventually be applied to them. Usually these resources do not undergo regular assessments. This study focuses on « ex quota » stocks fished by French fleets, in order to assess knowledge concerning them and analyse possible trends in their management. It was contracted with funding from the Maritime fisheries and aquaculture directorate. Thirty-four species in all were chosen with respect to their interest for French fleets and the probability of Europe's adopting supplementary management measures for them. Available data is sometimes patchy (or not...

Short-term storage and cryopreservation of turbot (Scophthalmus maximus) sperm ArchiMer
Chereguini, Olvido; Cal, Rosa Maria; Dreanno, Catherine; Ogier De Baulny, Bénédicte; Suquet, Marc; Maisse, Gérard.
Short-term storage over several days as well as cryopreservation of turbot (Scophthalmus maximus) sperm were studied. Two extenders, Ringer 200 and artificial seminal liquid (ASL2), are suggested for semen collection in order to avoid the deleterious effect of urine contamination, and for the purpose of short-term storage between 0 and 15 degrees C. Oxygen atmosphere is not suitable for turbot sperm storage. Turbot spermatozoa undergo cryopreservation with a high rate of success especially in a sucrose solution with 10% dimethyl sulfoxide (DMSO) and 10% egg yolk.

Siderophore production and probiotic effect of Vibrio sp. associated with turbot larvae, Scophthalmus maximus ArchiMer
Gatesoupe, Fj.
The proportion of vibrio E, dominant in healthy turbot larvae, was artificially increased in the rotifer enrichment medium. The maid effect of this vibrio enrichment was to improve the resistance of larval turbot challenged with a pathogenic strain of Vibrio splendidus, vibrio P. The improvement of the survival rates was significant with 1.5 x 10(6) CFU of vibrio E added per mi of enrichment medium, and it was still maximum at the hi hest dose tested (5 x 10(7) CFU.ml(-1); 80 +/- 2% of survivors after 48 h of challenge, versus 50 +/- 2 % for the control group). Further in vitro characterisation showed that vibrio E was able to grow in the presence of the chelating agents EDDHA and EDTA, while producing siderophore, as revealed by chrome azurol S dyeing....

Sperm features in turbot (Scophthalmus maximus): a comparison with other freshwater and marine fish species ArchiMer
Suquet, Marc; Billard, Roland; Cosson, Jacky; Dorange, Germaine; Chauvaud, Laurent; Mugnier, Chantal; Fauvel, Christian.
This paper presents some features of turbot sperm and compares them with data reported in the literature on other freshwater and marine fish. In captivity, turbot spermiation period lasts 6 months. When compared to other fish species, turbot males are characterized by low values of maximum gonadosomatic index (0.6-0.8%) and are poor sperm producers in terms of volume (0.2-2.2 ml), concentration (0.7-11.0 x 10(9) spermatozoa/ml) and total sperm number released per stripping (0.2-12.0 x 10(9) spermatozoa). Compared to most freshwater species, the duration of cell movement is long (1-17 min). The spermatozoon of turbot can be considered to be of a primitive type. A very high protein content (8.8 mg/ml) is found in the seminal fluid of turbot. Increasing...
